Strategy is determining where you are now, where you want to go, and how to get there. It’s the art of developing and implementing specific actions and decisions that will help a business to achieve goals and objectives as set out by the owners. Through the strategy process the overall direction of the company is set including opportunities and threats in the outside world and the internal strengths and weaknesses of the business. The aim of a business strategy is to ensure that the threats posed by the external environment are minimized and also to strengthen or minimize the effects of internal weaknesses. The opportunities and strengths are combined to ensure maximum productivity is achieved. The more the external environment changes the more opportunities there are likely for a company, but the more threats can be a problem if not prepared for. Without a business strategy, it would be difficult for an owner to realize the opportunities available. Strategic management in contrast with business policy is a technical approach having a five step model. Strategic management is a set of managerial decisions and actions that determine the long run performance of a company. Strategic management does not have to be just a part of the top level management. In fact, organizations should focus on strategic management at all levels of the management and the entire hierarchy of the company should be involved both in planning and implementation phases. This is what gets people hooked into buy-ins, followership, and implementation of the plan. So it means you just have to think outside the box of today and describe the world of the future. With the current situation of the economy the fact that some business establishments have stopped their operation and closed has caused big drop in small and large corporations. Having back up plan would be the idea should the original plan does not succeed businessmen could have alternative options or backed up plans for the continuity and success of the business. Since businessmen are knowledgeable of the business processes, they know the importance of having back up plans. Without a contingency plan, and should the original plan fail, businessmen and the business could experience a lot of trouble. The Mission statement is important because it describes the reason for the existence of an organization and what it hopes to achieve. While mission statements vary from organization to organization and represent the distinctness of each one, they all share similar components. Before writing a mission statement, leaders in the organization must have an idea of what is in store for the future. This vision is the foundation for the mission statement. Most statements include descriptions of the organization's target market, the geographic domain, their concern for survival, growth and profitability, the company philosophy, and the organization's desired public image. In addition, it is a tool that conveys the goal, organizational structure and strategy, legitimacy, values, participation and ownership among employees, leadership, responsibility to the community, ethical priorities, and commitment to the public and stakeholders. The organizational decision making process involves proper and efficient implementation of strategic plans and methods to achieve desired business objective. Often one difficulty facing an organization is that multiple divisions are involved in the overall decision making process. Making a decision can have different implications for each respective division. With the businesses of today facing shortened product lifecycles, organizations are facing intense competitive pressures to build market share to stay ahead of rivals. Process changes, the introduction of new and improved disruptive technologies are forcing companies to adapt new business goals and objectives in shortened timeframes, this alone places stresses on all organization. The Women Suffrage Movement (1840 -1920)

It would seem unbelievable that the women who lived before the 19th century were confined to their homes and actually had no freedom to choose the leaders that they wanted simply because they could not vote.This was not until the suffrage movement was formed to fight for the women's right to vote. During this time, men were still seen as more superior and women were only allowed to manage and participate in the domestic roles (Rossi 6).Only adult males were allowed to vote. Even though, they had to own property in order to qualify to vote. With changes in education and social conditions, women in the 19th century began to seek their rights and equality with men and top in their agenda was the right of women to vote just like their male counterparts.This saw the rise of the women suffrage movement of 1840 to 1920. The birth of this revolution is said to have occurred at a women rights meeting in Seneca Falls in New York. Women could see they were limited by their place in the society and in the Seneca Falls meeting the ‘Created Equal' campaign was launched (Rossi 8).Despite the many challenges that the women faced during this movement and an eighty years struggle, theirs became a won battle when the 19th amendment was signed in 1920 giving women the right to vote in national elections (Sochen 176).A suffragist known as Elizabeth Cady Stanton is the one that introduced women's sentiments at the Seneca Falls convection and wrote the plan for the feminist movement (Sochen 131). Later, together with Susan B. Anthony, she led women in the movement to fight for their rights of equality and their freedom to vote. Other significant leaders and suffrages included Lucretia Mott, Lucy Stone, Alice Paul, Virginia minor and Sojourner Truth.Elizabeth Stanton became the leading feminist philosopher guiding the American women towards the achievement of women's right. She was a determined woman rights activist who despite her responsibility as a wife and mother devoted as much time as she could in the movement (Salibury 131).Susan Anthony worked closely with Elizabeth and the two were said to be inseparable. Elizabeth is even said to have written Susan's speech at the Seneca Falls meeting. Susan was an acknowledged spokeswoman for the campaigns of the National American Women Suffrage Association (NAWSA) formed in 1878. She also wrote a magazine on women rights (Sochen 131). Alice Paul founded the founded the national women's party.The main objectives of this movement were for women to acquire their rights to vote equal to that of their male counterparts. The movement also sought to defend African American women who even after being granted franchise were not included in the fourteenth and fifteenth amends that gave them an equal opportunity to vote regardless of their race.The reasoning behind this movement is that no one should be denied a right to vote based on their gender or race. They wanted the fifteenth amend not only to include right to vote irrespective of race and color but to also include gender (Rossi 17). Other reasons were specialized for example in Utah; women believed that their rights to vote would bring an end to polygamy.The women suffrage movement managed to gain government support in granting voting rights to women. This they did through numerous campaigns and gaining the support of women all over America to support them. Many conferences and organizations to campaign for women rights to vote were organized in the leadership of Susan Anthony and Elizabeth Stanton.In 1871, about fifteen women registered and participated in the elections. Susan was arrested and in the following years, several more were arrested and harassed. The largest campaign was held in 1913 in Washington D. C when around five thousand women paraded showing signatures of one million women supporting suffrage for women (Rossi 35).Apart from national wide campaigns, women pursued suffrage in individual towns and gradually many of them were granted the rights to vote. Grants of suffrage were more successful in the west with the first suffrage succeeding in Wyoming in 1869. Other examples by years are Illinois; 1891, California; 1911, Arizona and Oregon; 1912 and Montana; 1914.The women of Utah were defeated after they voted for polygamy. The women suffrage was first introduced to the congress in 1915. The free entitlement provides access to education and care and the hours can be flexible over the week, all childcare provisions must use the EYFS and help young children achieve the five Every Child Matters outcomes. Describe what is meant by evidence-based practice and give examples of how this has influenced work with children in their early years. Evidence based practice is a framework of decisions made from effective information researched that influences practice and allows practitioners to apply their knowledge to a situation which allows them to make a well informed decisions on future actions. Professional practice requires these findings and research to be kept up-to-date and a consideration as to how these can be applied to settings. Sometimes, we need to be sure of what we find out before taking action as it might not be real or true. This is why it is so important for me to communicate and concerns or queries to my team. My everyday practice is influenced by what I have found out or learnt previously as a mother but also professionally through watching my nursery teacher and practitioners and learning as I go. It has been interesting to see that I am heading in the right track as a parent but excited to learn things I hadnt even thought of previously for my own children that I can not only use in the setting but also bring home for my children to benefit from. An example of how research has influenced working with children is The Effective Provision of Pre-School Education (EPPE) which is the first main study in the UK that focuses on the effectiveness of early years education and studies young childrens intellectual, social and behavioural development at age 3 to 7, collecting a wide range of information on more than 3000 children, their parents, their home environment and the pre-school they attended. Its findings found that children who had attended early years provision were more likely to have better cognitive, social and behaviour skills when they started formal education than those who had no early years provision, also confirming the value of early learning through play especially from low-income families. Key Elements of effective practice (KEEP) is another example. It stressed that effective learning in children is dependent on secure relationships. Learning through play and forming secure relationships are both key elements to the EYFS. With formal and non formal observation schedules and reflective practice, there is quality of care, learning and development and accountability as staff may require training and review of procedures and policies in order to keep up to date with new evidence in the provision of health and education. Explain what is meant by Diversity Diversity is the differences between individuals and groups of people in societies. These differences could be gender, ethnic origins, social cultural or religious background, family structure, disabilities, sexuality or appearance. Equality Equality means that individuals in society experience opportunities to achieve which are as good as the opportunities experienced by other people. Inclusion Inclusion is a process of identifying, understanding and breaking down barriers to participation and belonging. Explain the importance of anti-discriminatory/anti bias practice, giving examples of how it is applied in practice with children and carers The curriculum within settings should represent the different cultures of the children within it to promote anti-discriminatory and anti bias practice, as does my setting. We include positive images within the play areas for example in books and on posters which allows our children to view pictures of different races, disabilities, sex and age which promotes an anti-bias view of the world we live in. We also have children within the setting who have special educational needs which also brings it to life for all the children to encourage this acceptance into their lives. The setting creates an environment which is acceptable to all children regardless of their background, along with being able to make them feel welcomed. We ensure they are all valued and have access in every aspect of the provision. We also have had the pleasure of meeting practitioners who are from different countries which also helps the children relate to equality within their lives. Explain how the active participation of the children in decisions affecting their lives promotes the achievement of positive outcomes. Children need to actively participate in decisions that will affect their life. Children that are young need to make simple choices that enable them to find out their likes and dislikes not only for themselves but for us to then plan activities that meet their needs and challenge their abilities, it can be as simple as having a choice between an apple and a pear. This will enable them to express their needs and wishes. Letting the children make choices builds a child’s confidence, self esteem and their social skills. In our setting the children make choices all the time, this will be when a child makes a choice on what they want to do or who they want to do it with, these are simple choices that a child of pre-school and nursery age can make for themselves. This will enable the child to develop more and make choices later on in life that are more important such as, relationships, what course to take at college, whether to gain a qualification, what job they may undertake or even when to get married etc. These choices will all be big decisions that could affect the rest of their life. We as practitioners need to actively listen to the child’s choices and other ways that they may be able to communicate their wishes such as pointing, pictures, signing and any other non verbal means of communication to find out what the child or family actually want or need. We need to understand that children have voices and that they should be heard. Providing that they or others are not in any significant harm, their wishes should be listened to and followed. Examples of how children in our setting actively participate in their own choices include area of play, choice of activities during free flow time, choice of healthy food and drink available to them, choice of actions (good or bad, with intervention when necessary). Again, by giving them these choices it will help them to make small decisions in life and will enable them later in life to extend this to bigger decisions that affect their lives and will help future experiences. It also enables our children to learn to build up confidence, self-esteem and social skills by themselves and sometimes, with our guidance. Explain the importance of reviewing own practice as part of being an effective practitioner The quality of provision in any early years settings is dependent on the skills, attitudes, knowledge and experience of everyone who works there. Reflective practice is the key to quality improvement as it helps to identify the strengths and weaknesses of different aspects of a setting’s provision. Reflective practice is the term used to describe the way in which professionals evaluate their own work and consider ways of improving their work. It is important to do this, as each year a different set of children and parents join the setting. They may have different needs, expectations or interests. Activities, routines and polices may have to be changed or updated to accommodate the new families. Reflecting on practice will help to see where changes need to be made. Staff are developing all the time too. Their knowledge and skills may change due to extra training or new staff having new ideas. National standards and frameworks may also change so this may have an impact on how the setting is run. To reflect on my own practice I tend to critically question what I do and see whether it works or whether there is room for improvement by asking my team for feedback and advise. I am also new to childcare as a profession rather then parent so I am constantly taking things in and using support from my team as motivation to improve my own strengths and target my weaknesses. I tend to observe the reactions of children, parents and the team to help think about my effectiveness, but at the end of the day voicing any concerns is the best policy and my team will always come to me when needed. Especially in areas I may think are working well, I like to think about what skills and knowledge are helping me achieve this or whether I can become more efficient. In areas of weaknesses I always think of ways to improve and pick up hints and tips from my brilliant colleagues who are more then happy to advise. I may need more training in the future when I fell more experience is needed such as first aid etc. I find reflecting on my practise helpful as it gives me a clearer picture of what I actually do within my work role for my setting, and how I can make myself a valuable asset to my team. This itself gives me more confidence of my ability as practitioner and confidence to know that everybody can improve to provide the best care for all our children as I reach higher standards as I progress. Undertake a reflective analysis of own practice Being a reflective practitioner involves thinking about how you currently work and evaluating what you do in order to improve your practice. The reflective practitioner stands back, takes a balanced view and recognises what works well, but is also able to acknowledge what could be changed. To be a reflective practitioner you need to be self aware and able to look as objectively and honestly as possible at how you work with children, colleagues and parents. This is not easy, but taking a proactive role, reflecting on and analysing your own practice is far more rewarding than relying on someone in a more senior position to do this for you. Evaluating your own practice helps to put you in control of the changes that should be made, enables you to identify your professional development needs and will increase your confidence and feelings of job satisfaction. Reflective practice and self evaluation are fundamental to the Ofsted inspection process. By completing the Self Evaluation Form (SEF) managers and setting leaders are able to provide a snapshot of what happens in a setting. Through the SEF they can clearly identify the setting’s strengths and highlight what it does well. At the same time they can acknowledge any weaknesses in provision and plan the changes and improvements to be made. However, managers cannot achieve this alone. They are reliant on all practitioners in the setting taking responsibility for the quality of their individual practice and aiming for continuous improvement. As practitioner I want to be a good role model for the children in my setting by being enthusiastic to their efforts, positive and optimistic towards their abilities and good choices but also as I understand children are active learners I wish for them to be as enthusiastic as I am with their learning and development. I always aim to show a relaxed expression and warm tones in my voice that I can pitch higher as I express enthusiasm. I try to get the children eager to get involved especially in planned activities that are designed to assess the children individually as it is important for me to know each childs specific needs etc. I am aware that my relationships with the children, parents and colleagues are not only important for the setting environment but also for our children to understand how to treat others and how peers and teamwork is important in life. I treat others how I wish to be treated and respect all my team as individuals professionally as well as getting to know them and enjoying their company during work hours. It is a pleasure to work with a great group of people who are passionate about giving our children a great start in life which, in turn also gives me the motivation to thrive and provide the best care I can for my colleagues and children. I actively listen and am taking in lots of information and advise given verbally or through watching others. This is active learning and it is a cycle within the work setting. When I talk to my colleagues it is in a professional manner with interest and respect. I am also willing to help in any way I can to maintain an efficient smooth running setting. When I talk to the children in the setting I try to relate to them, coming down to their level with interest and listening to anything they have to say. They will always ask questions that I will answer as honestly as I can. I am always intrigued to know their likes and dislikes and by getting to know each individual child helps in this area. It is always nice to following up on their ideas by discussing every thing with the nursery teacher who will then advise. I try my hardest to make every parent feel welcome and at value what they tell me especially when it is about the importance of their individual child. It is important to maintain relationships with everyone as not only does it help me provide the best care I can but it also makes the place a very strong place to be as we are helping these children thrive and flourish to take these abilities with them through life. I try to help the setting by being proactive in solving problems as they arise but I will come to my team in times of need. I definitely need to continue to actively learn through my team but also establish a personal style and accept we are all different and all give a different benefit to the team. I started off with little confidence as a parent you never quiet know what boundaries you can cross with other children such as how stern to be when they make bad choices etc. Now I am much more confident and relaxed in my approach but I am always learning, every day I learn something new. I need to communicate and not be afraid to ask my nursery teacher if I misunderstand a task given to me. I made the mistake of setting out an extra activity that didnt help what the nursery teacher had planned as I assumed bowels were left out for it when in fact they were forgotten about and not put away. The nursery teacher was busy and so I tried to use my initiative but it was not a good decision. I should have waited to speak to my tutor and seek advice. This is what I shall do in the future. I tend to learn from my mistakes! Develop strategies to deal with areas of difficulty and challenges encountered in professional practice in early years settings Examples of areas of difficulty and challenges that may arise in the work setting between myself and parents of children include: Parents not collecting their child on time getting later and later Parents with outstanding fees Parents with a complaint e. g. they claim a child has hit their child Speaking to a parent about a concern you have about their child (you feel that the child has specific needs, and you are worried how they may accept/not accept it). Parent has been discriminating against another parent (possibly verbally to the other parents). These issues are not only very sensitive to each individual family but they all require handling with professionalism and respect as a parent myself you need to feel valued and listened to and made aware that the dilema will be dealt with promptly and effectively and in confidence. This is why having full and comprehensive policies and procedures within my setting is important and relevant at these times to follow. When speaking to parents, if they came to me with a concern or complaint I would listen, really listen, even if they start to shout, often they just want to say their piece. Then if they have cause to complain or bring something up I may be able to deal with it there and then by speaking in a professional manner, without making it personal. If I am able to resolve it, I will try, if not I would this to the nursery teacher as she is a very good active listener and always wants the best for our children, parents and team. It is very important to maintain relationships as we would hate to tarnish the settings name we really do enjoy our involvement and helping our children the best way we can. To me it doesnt matter how small the concern is its important for that child and their parents, and I want to assure those parents I take things very seriously in order to resolve issues. When a parent needs addressing for issues such as a concern about their child or fees are owed then the nursery teacher will deal with this matter by forms of letters or arranging a meeting to speak in confidence regarding the above. If a parent has come to me or the nursery teacher with a complaint about another parent I would have to have a quiet word or arrange a meeting to say It has been brought to my attention that you have been speaking about another parent unkindly/discriminatingly. They may interject here and say who said, they are lying? But I would just have to remember to say something such as I am not at liberty to say, but whether it is true or not I must remind you we are an equal opportunity setting who celebrates diversity and cultural beliefs. Remember to explain sensitively and nicely that you are duty bound to make sure all the children and families are treated equally, and you would do the same for their child, if you felt they needed your support. But currently it is the responsibility of the nursery teacher and so I would talk to her about any such issues.

Enzyme Kinetics: Lactate Dehydrogenase

Enzyme Kinetics: Lactate Dehydrogenase Question: What scientific question was this part of the practical designed to address? What is the effect of heat stability on two isoforms of the enzyme lactate dehydrogenase (LDH1 and LDH5) after pre-incubation (heat inactivation) of the enzyme samples at different temperatures? Approach: Outline and explain the experimental approach that was used to address the question. In order to determine the effect of the heat inactivation on LDH1 and LDH5, a colorimetric assay was carried out using different reaction mixtures prepared with LDH1 and LDH5. 16 test tubes were prepared each with a specific volume of LDH1 or LDH5. The same concentration of substrate solution containing lactate and NAD+ was added into each of the 16 tubes except for two tubes with the blank solution. Each of the enzyme samples were pre-incubated at 37 °Ãƒ Ã‚ ¡, 44 °Ãƒ Ã‚ ¡, 50 °Ãƒ Ã‚ ¡, 54 °Ãƒ Ã‚ ¡, 58 °Ãƒ Ã‚ ¡, 62 °Ãƒ Ã‚ ¡ and 66 °Ãƒ Ã‚ ¡. The reaction mixtures had to be mixed, incubated for 37 °Ãƒ Ã‚ ¡ and allowed to cool. This gives the lactate dehydrogenase enough time to work on the substrates under an optimum temperature. The lactate dehydrogenase catalyzes a reversible reaction by converting lactate and NAD+ to pyruvate, NADH and H+ or from pyruvate to lactate. The overall effect of the enzyme is that it transfers a hydride ion from lactate to NAD+ or from NADH to pyruvate. (Zhadin N et al 2008). The NADH produced in the LDH catalysed reaction has absorption properties (wavelength) that are greater than that of NAD+ which makes it useful for this experiment. When the colour reagent is added to the reaction solutions, NADH was used to form formazon (blue) according to the principle: Lactate + NAD+ → Pyruvate + NADH + H+ NADH + Phenazine methosulphate + H+ → NAD++ Reduced phenazine methosulphate. Reduced phenazine methosulphate + Tetrazolium → Formazan + Phenazine Methosulphate.The extent to which the enzymes have been heat inactivated will be reflected on the intensity of the formazon produced. The formazon was measured at 520nm in the spectrophotometer against the blank solutions for LDH1 and LDH5. The results obtained allowed us to compare how effective the LDH1 and LDH5 is at different pre-incubation temperatures and helps us to study the heat stability of the isoenzymes. Data: In figure 1 the absorbance begins to decline after around 44  °C in both enzyme isoforms. It is evident LDH5 has a higher absorbance than LDH1 at relatively low temperatures, but at 49 °C there is an overlap between the isoenzymes and after this point LDH5 continues to have a lower absorbance reading than LDH1. This shows the enzyme is losing effectiveness at catalysing the conversion reaction at temperatures higher than 44 °C suggesting this may be the enzyme’s optimum temperature at which it is most effective. With LDH1 there is a steady decrease in the enzyme activity of around 20-30% from 44 °C to 58 °C. After 58 °C there is a sharp drop in the effectiveness of the enzyme by around 60%. The decline may be due to the active site of the enzyme being denatured with exposure to the high pre-incubation temperature. The LDH5 begins with an increasing enzyme activity from 37 °C to 44 °C and after 44 °C there is a greater drop in enzyme activity of around 80%. The sharp drop in enzyme activity occurs much quicker in LDH5 than in LDH1 (figure 2), because there is a rapid loss of effectiveness when the temperature is raised above 50 °C. LDH5 also loses function at a lower temperature compared to LDH1; it becomes inefficient at 58 °C, where the curve remains consistent with a very low enzyme activity (0.31-0.62%). In addition, the LDH1 isoform demonstrates half maximal activity at 11.4 °C greater than that of LDH5 illustrating LDH1 is more tolerant to increasing temperatures. The results have shown that LDH5 has low heat stability and becomes inefficient at a lower temperature than LDH1. Discussion/Conclusions: The lactate dehydrogenase is characterised by its isoenzymes that are tetramers. The tetramers are made up of four monomers; H4 (heart) or M4 (muscle). The monomers are formed by a polypeptide chain of amino acids which consists of an active site. When there is lactate and NAD+ present in the solution, the peptide loop in lactate dehydrogenase blocks the access to the binding site after the substrates have bound; this aids the transition state of the reaction. The difference in heat stability between the two isoenzymes in the experiment can be due to the amino acid sequence of M and H subunits. The amino acid composition of M and H subunits consist of the same amino acids, however the difference is that they have varying numbers of each amino acid. LDH1 has a greater composition of valine, aspartic acid and methionine residues in comparison to LDH5 (Goldberg E.1972).The higher the methionine content in LDH1 means there is more sulphur atoms capable of forming strong covalent disulphide bridges, which can be why LDH1 is more heat stable as more energy is required to denature the isozyme compared to LDH5. Heat inactivation of the isoenzymes can affect the protein structure of lactate dehydrogenase. To form the primary structure, the -COOH group of one amino acid combines with the -NH of another to form a peptide bond. Within proteins there are regions that are arranged into alpha helices and beta pleated sheets. The bonds that hold these together are hydrogen bonds between lone pairs of oxygen and a hydrogen atom. The alpha helix is when the polypeptide is in a spiral form allowing more hydrogen bonds to form between the residues. Similarly, the beta pleated sheet consists of polypeptide chains running parallel or anti-parallel to each other. The way the polypeptide chain is arranged allows more hydrogen bonds to be formed. The two secondary structures fold up differently to form a 3D shape. There are various bonds that holds this shape together: ionic interactions between -COOH and -NH2 groups of different amino acids, hydrogen bonds between the side chains, van der Waals forces wher e fluctuating dipole in one of the groups of the amino acid can create a dipole in another which is used to keep the folded structure together, hydrophobic and disulphide bridges. A quaternary structure is formed by interaction of several polypeptide chains/subunits (LDH1 consists of 4 H subunits). In conclusion, the heat inactivation can interfere with the different interactions between the amino acids that make up the protein structures. The heat inactivation has shown to provide enough energy to overcome some of the covalent bonds to denature the enzymes. Since LDH1 is found to have a negative charge it may be associated with many ionic interactions which needs more heat energy is needed to overcome the strong bonds, hence why it takes a greater temperature (66 °Ãƒ Ã‚ ¡) for LDH1 to lose enzyme activity. The strong bonds holding the LDH1 together means the enzyme is not destroyed at 60 °Ãƒ Ã‚ ¡ however LDH5 is destroyed which is evident by its inability to function adequately after 55 °Ãƒ Ã‚ ¡. A coloured complex is formed using the mechanism in the previous experiment which produces blue formazon. The absorbance of the formazon was measured using the spectrophotometer; the absorbance measured indicates the enzyme activity. The results from this experiment will help to understand the effect of varying substrate concentration has on the enzyme activity and compare this between the two isoenzymes. To determine the Vmax and Km values more accurately the Lineweaver-Burk-plot is used. Discussion/Conclusions: As the substrate concentration is increased, the formation of the product will also rise but there is a point at which increasing the substrate concentration any further will become a limiting factor. The concentration at which this occurs is greater than 80mM which may decrease the enzyme activity in both LDH1 and LDH5. Since Km and Vmax is greater in LDH5 indicating the affinity of LDH5 for its substrate is much lower so a greater substrate concentration is needed to reach Vmax. The high Vmax for LDH5 means the saturated lactate dehydrogenase is converting more of the substrates into its products at high substrate concentrations. As a result LDH1 will be a more ideal isoenzyme than LDH5. In this experiment we observed the catalysis of lactate dehydrogenase from lactate to pyruvate. The pyruvate produced has the ability to enter the mitochondria, where pyruvate dehydrogenase links the glycolysis metabolic reaction to the citric acid cycle. In a process of pyruvate decarboxylation, the pyruvate is converted into acetyl-CoA, which releases energy by converting NAD+ to NADH and H+. Pyruvate has the potential to either enter the mitochondria to become acetyl-CoA or become lactate. The Cori cycle is involved in the conversion of lactate to pyruvate. When lactate is produced in the muscle, it is released into the bloodstream and transported to the liver. In the liver the lactate is first converted to pyruvate by lactate dehydrogenase and pyruvate is converted to glucose through gluconeogeneis. Lactate dehydrogenase becomes activated when there is a high concentration of its substrates, NAD+ and lactate. For instance when the skeletal muscle containing LDH5 have high demands for energy this means the large production of NADH and pyruvate from high substrate levels will not be metabolised by pyruvate dehydrogenase. The LDH5 (M form) is more specialised to convert pyruvate to lactate and ideal for anaerobic activities. Whereas the LDH1 form is more favourable for the conversion of lactate to pyruvate, LDH1 is predominantly found in the heart where there is an aerobic environment. This means despite increasing the substrate concentration, the LDH5 may not become as efficient in converting lactate to pyruvate as LDH1. The results reflect this as LDH1 has a generally greater enzyme activity with an increasing substrate solution compared to LDH5 (see table of results). In conclusion, the results obtained from the analysis of data on excel may not be highly accurate since most of the values have been rounded and it does not take into account any possible practical or human errors. Whatever amount of work has been done in class, a teacher is to give the students the same amount of homework each day, between one and two hours a night. III. A teacher is not to make any exceptions for his tardy work policy no matter what the excuse. The punishment for someone who had a big game or whose house was burned down or whose house was buried under ten feet of snow shall be the same as someone who has no reasonable excuse.
